Rabies is an infectious viral disease without any cure. Autophagy considered as a method for reducing the damage to cell and its organelles. In some virus-infected cells, autophagy plays antiviral effect, while it acts in opposite manner other cells. Beclin1 (Atg6/Vps30) main regulator of autophagy. The present study aims build recombinant beclin1 stimulating evaluating effects mice infected with street rabies virus. First, pIRES2-EGFP-beclin1 vector was made by PCR. Then, virus (SRABV), pIRES2-EGFP were injected into brain mice. addition, sacrificed samples taken PCR, FFA, real-time Tunnel assay. accuracy pIRES2-EGFP-beclin 1 confirmed colony PCR digestion restriction enzymes. vitro expression tested gene sequencing, SDS-PAGE, Western blot. Further, function vivo investigated transfecting cells CHO mouse brain. result beclin1sequence blasting existing sequences GenBank database revealed high levels homology mutation. correct appropriate role can be used researchers research purposes.
